A systematic survey of LU domain-containing proteins reveals a novel human gene, LY6A, which encodes

摘要: The Ly-6 and uPAR (LU) domain-containing proteins represent a large family of cell-surface markers. In particular, mouse Ly-6A/Sca-1 is a widely used marker for various stem cells; however, its human ortholog is missing. In this study, based on a systematic survey and comparative genomic study of mouse and human LU domain-containing proteins, we identified a previously unannotated human gene encoding the candidate ortholog of mouse Ly-6A/Sca-1. This gene, hereby named LY6A, reversely overlaps with a lncRNA gene in the majority of exonic sequences. We found that LY6A is aberrantly expressed in pituitary tumors, but not in normal pituitary tissues, and may contribute to tumorigenesis. Similar to mouse Ly-6A/Sca-1, human LY6A is also upregulated by interferon, suggesting a conserved transcriptional regulatory mechanism between humans and mice. We cloned the full-length LY6A cDNA, whose encoded protein sequence, domain architecture, and exon‒intron structures are all well conserved with mouse Ly-6A/Sca-1. Ectopic expression of the LY6A protein in cells demonstrates that it acts the same as mouse Ly-6A/Sca-1 in their processing and glycosylphosphatidylinositol anchoring to the cell membrane. Collectively, these studies unveil a novel human gene encoding a candidate biomarker and provide an interesting model gene for studying gene regulatory and evolutionary mechanisms.

FERM domain-containing protein FRMD6 activates the mTOR signaling pathway and promotes lung cancer progression

摘要: FRMD6, a member of the 4.1 ezrin–radixin–moesin domain-containing protein family, has been reported to inhibit tumor progression in multiple cancers. Here, we demonstrate the involvement of FRMD6 in lung cancer progression. We find that FRMD6 is overexpressed in lung cancer tissues relative to in normal lung tissues. In addition, the enhanced expression of FRMD6 is associated with poor outcomes in patients with lung squamous cell carcinoma (n = 75, P = 0.0054) and lung adenocarcinoma (n = 94, P = 0.0330). Cell migration and proliferation in vitro and tumor formation in vivo are promoted by FRMD6 but are suppressed by the depletion of FRMD6. Mechanistically, FRMD6 interacts and colocalizes with mTOR and S6K, which are the key molecules of the mTOR signaling pathway. FRMD6 markedly enhances the interaction between mTOR and S6K, subsequently increasing the levels of endogenous pS6K and downstream pS6 in lung cancer cells. Furthermore, knocking out FRMD6 inhibits the activation of the mTOR signaling pathway in Frmd6−/− gene KO MEFs and mice. Altogether, our results show that FRMD6 contributes to lung cancer progression by activating the mTOR signaling pathway.

摘要:

PTPN11, which encodes tyrosine phosphatase Shp2, is a critical gene mediating cellular responses to hormones and cytokines. Against original prediction as tumor suppressor for tyrosine phosphatases, PTPN11 was first identified as a proto-oncogene because activating mutations of this gene are associated with leukemogenesis. However, most recent experimental data suggest PTPN11/Shp2 acting as a tumor suppressor in hepatocarcinogenesis. This review focuses on the tumor-promoting or suppressing roles of the gene PTPN11/Shp2 in different cell types.

摘要: Familial acne inversa (AI) is an autoinflammatory disorder that affects hair follicles and is caused by loss-of-function mutations in -secretase component genes. We and other researchers showed that ( ) is the most frequently mutated gene in familial AI. In this study, we generated a keratin 5-Cre-driven epidermis-specific conditional knockout mutant in mice. We determined that this mutant recapitulated the major phenotypes of AI, including hyperkeratosis of hair follicles and inflammation. In mice, the IL-36a expression level markedly increased starting from postnatal day 0 (P0), and this increase occurred much earlier than those of TNF- , IL-23A, IL-1 and TLR4. RNA-Seq analysis indicated that Sprr2d, a member of the small proline-rich protein 2 family, in the skin tissues of the mice was also upregulated on P0. Quantitative reverse-transcription polymerase chain reaction showed that other genes had a similar expression pattern. Our findings suggested that IL-36a might be a key inflammatory cytokine in the pathophysiology of AI and involved in the malfunction of the skin barrier in the pathogenesis of AI.

PROTECTIVE ROLES OF D1 PROTEIN TURNOVER AND THE XANTHOPHYLL CYCLE IN TOMATO (

摘要: D1 protein turnover and the xanthophyll cycle (XC) are important photoprotective mechanisms in plants that operate under adverse conditions. Here, streptomycin sulfate (SM) and dithiothreitol (DTT) were used in tomato plants as inhibitors of D1 protein turnover and XC to elucidate their photoprotective impacts under sub-high temperature and high light conditions (HH, 35°C, 1000 µmol·m ·s ). SM and DTT treatments significantly reduced the net photosynthetic rate, apparent quantum efficiency, maximum photochemical efficiency, and potential activity of photosystem II, leading to photoinhibition and a decline in plant biomass under HH. The increase in reactive oxygen species levels resulted in thylakoid membrane lipid peroxidation. In addition, there were increased non-photochemical quenching and decreased chlorophyll pigments in SM and DTT application, causing an inhibition of D1 protein production at both transcriptional and translational levels. Overall, inhibition of D1 turnover caused greater photoinhibition than XC inhibition. Additionally, the recovery levels of most photosynthesis indicators in DTT-treated plants were higher than in SM-treated plants. These findings support the view that D1 turnover has a more important role than XC in photoprotection in tomato under HH conditions.

摘要: In this paper, the natural convection in a square enclosure with a rectangular heated cylinder is investigated via the lattice Boltzmann method. A detailed study is conducted on the effect of the cylinder width and the Rayleigh number on the fluid flow and heat transfer. The flow structures and heat transfer patterns are classified into eight buoyant regimes, i.e., four steady regimes, two periodic regimes, one multiple periodic regime, and one chaos regime, two of which are reported for the first time.

Treatment of soybean wastewater by a wild strain Rhodobacter sphaeroides and to produce protein under

摘要: The conventional treatment method of soybean wastewater is expensive and generates waste sludge that requires further handling. Purple nonsulfur bacteria (PNSB) wastewater treatment is a clean technology and can generate single cell protein while degrading pollutants. A wild strain of PNSB, Z08, was isolated from local soil and was used to treat soybean wastewater. To develop a cost-effective process, the work was performed under natural conditions without artificial light, aeration, nutrients addition, or pH and temperature adjustment. The results showed that the wild strain Z08 could grow well under natural conditions. The growth curve showed two quick-growth periods and a turning point. The Z08 treatment of soybean wastewater was zero order reaction and COD reduction was 96% after 10 d. The major byproducts of the process were C2-C5 organic acids, predominantly butyric acid. No alcohol was found in the effluent. The initial COD/bacterial-mass ratio (F/M) had a significant effect on soybean wastewater treatment efficiency. When the initial F/M was lower than 10 mg-COD/mg-bacteria, a sufficient amount of time to achieve 90% of COD reduction was only three days. The Z08 biomass yield was 0.28g·g, and the bacterial protein content was 52%.

Development and clinical trial of a novel bioactive bone cement

摘要: Strontium (Sr) and related compounds have become more attractive in the prevention and treatment of osteoporosis. Previously, we developed a novel bioactive bone cement which is mainly composed of strontium-containing hydroxyapatite (Sr-HA) filler and bisphenol A diglycidylether dimethacrylate (Bis-GMA) resin. This bone cement is superior to conventional polymethylmethacrylate (PMMA) bone cement in bioactivity, biocompatibility, and osseointegration. It also has shown sufficient mechanical strength properties for its use in percutaneous vertebroplasty (PVP) and total hip replacement (THR). In this paper, we review the and clinical evidence for the effectiveness of this bioactive bone cement.

Moving target detection in the cepstrum domain for passive coherent location (PCL) radar

摘要: A cepstrum moving target detection (CEPMTD) algorithm based on cepstrum techniques is proposed for passive coherent location (PCL) radar systems. The primary cepstrum techniques are of great success in recognizing the arrival times of static target echoes. To estimate the Doppler frequencies of moving targets, we divide the radar data into a large number of segments, and reformat these segments into a detection matrix. Applying the cepstrum and the Fourier transform to the fast and slow time dimensions respectively, we can obtain the range information and Doppler information of the moving targets. Based on the CEPMTD outlined above, an improved CEPMTD algorithm is proposed to improve the detection performance. Theoretical analyses show that only the target’s peak can be coherently added. The performance of the improved CEPMTD is initially validated by simulations, and then by experiments. The simulation results show that the detection performance of the improved CEPMTD algorithm is 13.3 dB better than that of the CEPMTD algorithm and 6.4 dB better than that of the classical detection algorithm based on the radar cross ambiguity function (CAF). The experiment results show that the detection performance of the improved CEPMTD algorithm is 1.63 dB better than that of the radar CAF.

Epidemiological study of a von Hippel-Lindau family in northwest China

von Hippel-Lindau (VHL) disease is a rare, inherited neoplastic disease characterized by hemangioblastomas (HBL) of the central nervous system (CNS), retinal angiomas, renal cell carcinomas (RCC), pancreatic endocrine tumors (PETs), pheochromocytomas, paragangliomas, and visceral cysts. We encountered a large VHL family in northwest China and conducted a systematic screening of the family members based on their epidemiological and clinical characteristics. A self-designed questionnaire was used to collect the general sociodemographic and health information of the family members. For the preliminary family screening, physical examination and abdomen B ultrasonography were performed. The suspected patients were subjected to cranial computerized tomography and fundus examination. The clinical data of the patients with confirmed VHL disease were collected from hospital records. A total of 63 lineal descendants in six generations were observed in the family (generations O, A, B, C, D, E), including 9 dead suspected cases (6 males, 3 females) and 10 living cases (2 males, 8 females). Among the 10 living cases, 4, 2, 1, 3, 4, 8, and 2 manifested HBLs of the CNS, PETs, RCC, pancreatic cysts, renal cysts, pheochromocytomas (4 hemi and 4 bilateral), and paragangliomas, respectively. Data showed that the morbidity of VHL disease in generation C was lower than that in generation B, but the age of onset was younger. This study is the first to report VHL disease in northwest China and VHL-associated PET cases in Chinese. Therefore, follow-up checkups of the family should be focused on younger generations. Proper family screening protocols should be followed for the treatment of patients with VHL disease.

Hepatitis B virus X protein upregulates tumor necrosis factor-α expression of rat mesangial cell

摘要: Hepatitis B virus X protein (HBx), a 17-kd protein encoded by X gene of hepatitis B virus (HBV), has been shown to function as a transcriptional trans-activator of a variety of viral and cellular promoter/enhancer elements. The aim of the study is to investigate the extracellular regulated protein kinases (ERKs) pathway of HBx on glomerular mesangial cell (GMC) proliferation and tumor necrosis factor-α (TNF-α) expression. The HBV X gene was amplified by polymerase chain reaction (PCR), inserted into the eukaryotic expression vector pCI-neo and confirmed by restriction endonuclease digestion and sequence analysis. PCI-neo containing HBV X gene (pCI-neo-X) was then transfected into cultured GMC line via liposome. GMC proliferation, TNF-α and its mRNA expression were compared in the condition of with or without U0126 in culture media. HBx, ERK and p-ERK expression in GMCs was assessed by Western blotting. TNF-α mRNA expression was assessed by semi-quantitative reverse transcription-PCR (RT-PCR). TNF-α level in supernatants was measured by ELISA. GMC proliferation was detected by 3-(4,5-dimethylthiazol-2yl)-2,5-diphenyltetrazolium bromide (MTT) kit. The results showed that HBx expression was found in transfected GMCs and became prominent at 36th and 48th h after transfection whether with or without U0126 in culture media. TNF-α mRNA expression was significantly decreased in U0126 group compared with U0126-free group. TNF-α levels in supernatants in PCI-neo-X transfection without U0126 group were (189.0±18.1) and (172.3±24.3) pg/mL at 36th and 48th h after transfection, respectively. In contrast, TNF-α levels in supernatants with U0126 were (65.6±11.6) and (84.0±24.6) pg/mL at 36th and 48th h, respectively. The TNF-α levels in the latter groups were significantly lower than those in the former groups (<0.05). GMCs proliferation was also lower in added U0126 group at 36th and 48th h after transfection. From above, we can conclude that HBx could induce GMC proliferation and increase TNF-α mRNA expression and its protein production. HBx upregulates TNF-α expression and induces cell proliferation of GMC line partly through ERK signal transduction pathway.

Time-domain and frequency-domain approaches to identification of bridge flutter derivatives

摘要: Flutter derivatives are essential for flutter analysis of long-span bridges, and they are generally identified from the vibration testing data of a sectional model suspended in a wind tunnel. Making use of the forced vibration testing data of three sectional models, namely, a thin-plate model, a nearly streamlined model, and a bluff-body model, a comparative study was made to identify the flutter derivatives of each model by using a time-domain method and a frequency-domain method. It was shown that all the flutter derivatives of the thin-plate model identified with the frequency-domain method and time-domain method, respectively, agree very well. Moreover, some of the flutter derivatives of each of the other two models identified with the two methods deviate to some extent. More precisely, the frequency-domain method usually results in smooth curves of the flutter derivatives. The formulation of time-domain method makes the identification results of flutter derivatives relatively sensitive to the signal phase lag between vibration state vector and aerodynamic forces and also prone to be disturbed by noise and nonlinearity.
